ABSTRACT

Objective To explore the value of improved CT perfusion in delineation of brain arteriovenous malformation(AVM) target for stereotactic radiosurgery.Methods 22 patients diagnosed with AVM by DSA were included in this study.14 cases of AVM were detected from initial symptoms of intracereb.ral hemorrhage,of which 4 cases were given immediate intracranial hematoma evacuation,then in 3 cases postoperative embolization was performed,and other 10 cases received conservative treatment including 3 cases treated by embolization.8 cases of AVM were detected from initial symptoms of epilepsy or headache,without surgery or embolization treatment.In all patients,the improved CT perfusion and MRA examinations were performed before treatment to evaluate the diagnostic efficacy of different methods in AVM.Results The interference rates of hemorrhage and granulation tissue on MRA images were 27.3 % and 54.5 %,respectively,while those on enhanced CT and improved CT perfusion images were 0 %.The interference rate of embolization material on enhanced CT and improved CT perfusion images was 27.3%,while that on MRA images were 0%.The contrast-enhancement rates of MRA,CT and improved CT perfusion images were 4 5.5 %,5 4.5 % and 7 2.7 %,respectively.Conclusion Improved CT perfusion technique is helpful in delineation of brain AVM target for stereotactic radiosurgery in patients with AVM combined with intracerebral hemorrhage or postoperative patients.

ABSTRACT

Bone morphogenetic protein-4(BMP4) is one of BMPs member,it not only can play an important role in cell proliferation and invasion,but also play an important role in cell differentiation,apoptosis and angiogenesis.A study showed BMP4 in malignant tumor can inhibit the growth of tumor cells,and also can induce tumor cell invasion,metastasis and epithelial mesenchymal transformation,and SMAD is the classic signaling pathways.BMP4 is expected to become a potential therapeutic targets and in evaluating the prognosis,immune therapy in malignant tumor.In this paper,the structure and function of BMP4 and its mechanism in tumor were reviewed.

ABSTRACT

OBJECTIVE:To establish a method for the simultaneous determination of curdione,germacrone and furanodiene of zedoray turmeric oil in Ruxiankang injection. METHODS:HPLC was performed on the column of Zorbax SB-C18 with mobile phase of acetonitrile-water (gradient elution) at a flow rate of 1.0 ml/min,detection wavelength was 216 nm,column temperature was 30℃,and the injection volume was 10 μl. RESULTS:The linear range was 60-480 μg/ml for curdione(r=0.999 0),40-320 μg/ml for germacrone(r=0.999 0)and 40-320 μg/ml(r=0.999 0);RSDs of precision,stability and reproducibility tests were lower than 2.0%;recoveries were 95.21%-99.89%(RSD=1.6%,n=6),102.33%-104.89%(RSD=1.0%,n=6)and 97.38%-99.06%(RSD=0.7%,n=6),respectively. CONCLUSIONS:The method is simple and accurate,and can be used for simultaneous contents deter-mination of curdione,germacrone and furanodiene of zedoray turmeric oil in Ruxiankang injection.

ABSTRACT

Objective To establish an HPLC method for determining the content of swertiamarin in Gantaishu capsules. Methods Using a Waters Symmetry C18 column (4.6 mm×250 mm, 5μm), the mobile phase was acetonitrile-0.02%phosphoric acid, with flow rate of 1.0 mL/min. The detection wavelength was at 250 nm, the column temperature was room temperature. Results The content of swertiamarin showed a good linear relationship in the range of 0.373 556-2.988 448 μg, r 2=1.000 0. The average recovery was 106.81%, and RSD was 3.7%. Conclusion This method is fast, accurate and simple with good reproducibility, which can be applied to the quality control of Gantaishu capsules.

ABSTRACT

Objective To establish a method for the determination of berberine hydrochloride and phellodendrine in Lishukang Capsules by HPLC. Methods Waters Symmetry C18 column (4.6 mm× 250 mm, 5μm) was used, the mobile phase was acetonitrile-0.1%phosphoric acid (0.1 g sodidum dodecyl sulfonate in 100 mL water), with the flow rate of 1 mL/min by gradient elution. The detection wavelength was set at 284 nm. Results Berberine hydrochloride showed good linear relationship (r 2=0.999 1) in the range of 403.4-10 084.7 μg, the average recovery rate was 99.8%(RSD=1.7%). Phellodendrine showed good linear relationship (r 2=0.999 2) in the range of 169.137 6-4228.44 μg, the average recovery rate was 95.5%(RSD=3.2%). Conclusion The method is specific, accurate and convenient to be used for the quality control of Lishukang Capsules.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the incidence of constipation after stroke and explore the relationship between post-stroke constipation and brain injury.</p><p><b>METHODS</b>Using a self-designed questionnaire, we collected the general information of 723 inpatients from 10 hospitals in Guangzhou, including the general demographic information, related factors of brain injury, defecation and previous history.</p><p><b>RESULTS</b>The total incidence of post-stroke constipation was 34.6% in these patients. The incidence of constipation was 31.0% in ischemic stroke patients, 44.8% in hemorrhagic stroke patients, and 47.4% in patients with cerebral hemorrhage and infarction, showing a significant difference between stroke types (χ(2)=12.369, P=0.002). In terms of stages following stroke, the incidence was 41.6% in the acute phase, 31.5% in the recovery phase, and 22.6% in the sequelae phase. In light of lesion locations, the incidence was significantly higher in patients with basal ganglia involvement than in those without (P<0.001). Logistic multivariate analyses showed that stroke type, post-stroke stage, lesion number and basal ganglia involvement were significantly associated with the incidence of constipation.</p><p><b>CONCLUSION</b>Constipation often occurs in the acute stage following stroke (especially hemorrhagic stroke), and the incidence is higher in patients with basal ganglia involvement.</p>

ABSTRACT

Objective To explore the effects of needle retractable syringe.Methods 400 patients undergoing injection treatment were randomly divided into the experiment group and the control group in equal number.The conventional syringe was used in the control group and the needle retractable syringe was adopted.The pains in the patients,time for puncturing and incidence of puncturing-induced injuries were compared between two groups.Result The pains,time for puncturing and incidence of puncturing-induced injuries in the observation group were all significantly lower than those of the control group(all P < 0.05). Conclusion Needle retractable syringes may help to alleviate the pain,reduce the workload of nurses and lower the incidence of needle stick injuries.

Objective To probe into the influencing factors of selecting the intravenous vascular access device for tumor patients undergoing intravenous therapy? Methods With convenience sampling,376 tumor patients in oncology department from 3 hospitals of class A hospitals in Guangzhou were selected for the survey including such 6 categories with 41 items as general data, patient’s willingness,punctured blood vessels,use of vascular access and clinical conditions? Results In 124 patients,peripheral venous catheter was used (33?0%)? Central venous catheter was used in 252 patients (67?0%),Multiple linear regression analysis showed that way for paying the expense,vascular conditions for venous punctuation,phased diagnosis,decision of the doctors,and prospected duration for detaining catheter significant influencing factors? Conclusions The state should raise their medical allowances for those with lower incomes? The administrative should enhance the training to doctors and nurses at grass levels,promote the communication between medical workers and patients and improve their recognition of transfusion devices so that they can make a right choice?

ABSTRACT

Objective To understand the status of cognitive dysfunction among new onset constipation after stroke and to explore the relationship between them.Methods With a self-designed questionnaire,the general information of 723 inpatients from 10 hospitals in Guangzhou were collected,including general demographic information,stroke situation,defecation situation and Mini-mental State Examination (MMSE)etc.Results The incidence of new-onset constipation,cognitive dysfunction(PSCD)after stroke was respectively 34.6% and 62.4%.Among whom 31.0% in ischemic stroke,44.8% in hemorrhagic stroke,47.4% in cerebral hemorrhage and infarction.There was statistical significance between stroke type and constipation,and ischemic stroke had statistical significance compared to other two types.During the acute phase reached 41.6%,the recovery phase 31.5% and the sequelae phase 22.6%.The rates of constipation patients with cognitive dysfunction were higher than patients without cognitive dysfunction,and the difference in directional force,memory,attention and calculation force,reading and expression was statistically significant respectively,among which expression was the most relevant.The difference in the incidence of constipation among different cognitive dysfunction groups was statistically significant,and they were positively correlated,compared with normal cognitive function,the light,medium and severe cognitive dysfunction was 1.519,2.879,3.064 times respectively.Conclusions Medical staff need to be alerted to the possibility that patients with impared cognitive function may be at greater risk of constipation.The preventive and treatment measures should be emphasized in order to improve patients' cognitive function.

ABSTRACT

The effects of a novel immunosuppressive agent FTY720 on proliferation inhibition and apoptosis of acute leukemia cell lines HL-60 and U937, and the role of extracelluar regulated protein kinase (ERK) in the course of proliferation inhibition and apoptosis induced by FTY720 were studied. The proliferation inhibition rate of HL-60 and U937 cells by various concentrations of FTY720 was detected by MTT assay. Cell apoptosis was detected by DNA fragment analysis and flow cytometry. The phosphorylated ERK1/2 protein expression was observed by Western blotting. The change of intracellular distribution of ERK1/2 protein was identified by SP immunohistochemical staining. The results showed that FTY720 could inhibit the growth of HL-60 and U937 cells effectively in a dose-dependent manner. After incubation with FTY720 for 24 h, apoptosis was observed in HL-60 and U937 cells. The intracellular expression of phosphorylated ERK1/2 protein was also down-regulated and the distribution of ERK1/2 protein in cell nuclear was reduced during FTY720-induced apoptosis. So, that FTY720 inhibited ERK1/2 phosphorylation might mediate the role of FTY720-induced apoptosis and proliferation inhibition of leukemia cells.

ABSTRACT

The expression and functional activity of multiple drug resistance (MDR1) gene in human normal bone marrow CD34+ cells was observed. Human normal bone marrow CD34+ cells were enriched with magnetic cell sorting (MACS) system, and then liposome-mediated MDR1 gene was transferred into bone marrow CD34+ cells. Fluorescence-activated cell sorter was used to evaluate the expression and functional activity of P-glycoprotein (P-gp) encoded by MDR1 gene. It was found that the purity of bone marrow CD34+ cells was approximately (91 +/- 4.56)% and recovery rate was (72.3 +/- 2.36)% by MACS. The expression of P-gp in the transfected CD34+ cells was obviously higher than that in non-transfected CD34+ cells. The amount of P-gp in non-transfected CD34+ cells was (11.2 +/- 2.2)%, but increased to (23.6 +/- 2.34)% 48 h after gene transfection (P<0.01). The amount of P-gp was gradually decreased to the basic level one week later. The accumulation and extrusion assays showed that the overexpression of P-gp could efflux Rh-123 out of cells and there was low fluorescence within the transfected cells. The functional activity of P-gp could be inhibited by 10 microg/ml verapamil. It was suggested that the transient and highly effective expression and functional activity of P-gp could be obtained by liposome-mediated MRD1 transferring into human normal bone marrow CD34+ cells.

ABSTRACT

In order to investigate the regulative function of telomerase and phosphorylated (activated) extracellular regulated protein kinase (ERK) 1 and 2 in the leukemic cell lines HL-60 and K562 proliferation inhibition and apoptosis, three chemotherapeutic drugs Harringtonine (HRT), Vincristine (VCR) and Etoposide (Vp16) were selected as inducers. The proliferation inhibition rate was detected by MTT method, the cell cycle and cell apoptosis was analyzed by flow cytometry and the telomerase activity was detected by the telomeric repeat amplification protocol (TRAP) assay and bioluminescence analysis method. The phosphorylated ERK1/2 protein expression was detected by western blot method. The results showed that HRT, VCR and Vp16 could inhibit cell proliferation, induce apoptosis, inhibit telomerase activity and down-regulate the protein expression of phosphorylated ERK. It was suggested that ERK signal transduction pathway was involved in the down-regulation of telomerase activity and the onset of apoptosis in the leukemic cells treated by HRT, VCR and Vp16.

ABSTRACT

To explore the effect of NF-κB on bcl-x gene transcription in extended drug resistance leukemia cell line HL-60/E6, drug-resistant subline HL-60/E6 was derived by intermittently exposing HL-60 cells to 6 ng/ml epirubicin. Indirect immunofluorescence was used to demonstrate the location of NF-κB-RelA in HL-60/E6 cells. FCM analysis and RT-PCR were used to detect the efficiency of liposome-mediated ODN transfection and the change of bcl-xL mRNA levels after 5 μmol/L phosphorothioate (PS)-derivatized antisense (AS) oligodeoxynucleotide (ODN) directed to RelA was transferred into HL-60/E6 cells. The results showed that RelA remained persistently active and located at the nuclei of HL-60/E6 cells,but in the cytoplasm of HL-60 cells, the efficiency of liposome-mediated ODN transfection was significantly higher than that of null ODN (P<0.01 in 4 h, 6 h, 12 h, 24 h). Exposure of HL-60/E6 cells to 5 μmol/L AS-PS-ODN directed to RelA led to a maximal 40 % decline of bcl-xL mRNA levels within 8 h. The inhibition rate of bcl-xL mRNA was (15±1.79) %, (28±2.34) %, (40±3.47) %, (20±1.54) % in 4 h, 6 h, 8 h, 15 h, respectively, but it was less than 15 % in control group. It was concluded that NF-κB was involved in regulating bcl-x transcription. It was suggested that NF-κB was an important factor for drug resistance in leukemia cells.

ABSTRACT

AIM: To explore whether inhibition of NF-?B by antioxidant pvrrolidine dithiocarbamate (PDTC) sensitizes leukemia cells to cytotoxic drugs and its mechanism. METHODS: The indirect immunofluorescence method and electrophoretic mobility shift assay (EMSA) were used to measure the activation of NF-?B. The apoptotic cells were evaluated by flow cytometry (FCM) and the in vitro growth inhibitory effect was performed using a MTT assay. RESULTS: EMSA showed that NF-?B was activated by daunorubicin (DNR), VP-16 and then was inhibited by PDTC in a dose-dependent manner. NF-?B activation was further verified because of subunit RelA of NF-?B locating in the nuclei. FCM analysis showed that apoptotic index of HL-60 cells was up to (8.97?0.81)%, (16.01?1.06)%, (22.96?1.33)% from (5.34?0.62)%, (10.16?0.42)%, (17.32?1.15)% after exposure of HL-60 cells to 2.5-10 mg/L VP-16 combined with PDTC.
